چکیده

Over the past 20 years, there has been considerable interest in the elucidation of factors that mediate angiogenesis. Early efforts, including Michaelson’s description of “Factor X” in mediating retinal neovascularization and Folkman’s attempts at isolating “tumor angiogenesis factor,” have blossomed into a plethora of investigations identifying regulators, both positive and negative, of blood vessel formation.1,2 These efforts led to the identification of factors such as vascular endothelial growth factor, acidand basic-fibroblast growth factor, angiogenin, and angiopoietins, to name a few.3–5 Subsequent detailed investigations have revealed that finely tuned and precisely coordinated expression of these factors is critical for angiogenesis during both development and adult life. Failure to coordinate expression has significant implications, as excess angiogenesis can contribute to tumor growth and inflammatory disorders, whereas insufficient angiogenesis can contribute to ischemic heart disease and preeclampsia.6

Fibroblast growth factor signaling pathway in endothelial cells is activated by BMPER to promote angiogenesis.

OBJECTIVE Previously, we have identified bone morphogenetic protein endothelial cell precursor-derived regulator (BMPER) to increase the angiogenic activity of endothelial cells in a concentration-dependent manner. In this project, we now investigate how BMPER acts in concert with key molecules of angiogenesis to promote blood vessel formation. APPROACH AND RESULTS To assess the effect of BMP...

BMPER is an endothelial cell regulator and controls bone morphogenetic protein-4-dependent angiogenesis.

Bone morphogenetic proteins (BMPs) are involved in embryonic and adult blood vessel formation in health and disease. BMPER (BMP endothelial cell precursor-derived regulator) is a differentially expressed protein in embryonic endothelial precursor cells. In earlier work, we found that BMPER interacts with BMPs and when overexpressed antagonizes their function in embryonic axis formation. In cont...

LRP1-dependent endocytic mechanism governs the signaling output of the bmp system in endothelial cells and in angiogenesis.

RATIONALE Among the extracellular modulators of Bmp (bone morphogenetic protein) signaling, Bmper (Bmp endothelial cell precursor-derived regulator) both enhances and inhibits Bmp signaling. Recently we found that Bmper modulates Bmp4 activity via a concentration-dependent, endocytic trap-and-sink mechanism. OBJECTIVE To investigate the molecular mechanisms required for endocytosis of the Bmp...

Bone morphogenetic protein endothelial cell precursor-derived regulator regulates retinal angiogenesis in vivo in a mouse model of oxygen-induced retinopathy.

OBJECTIVE Bone morphogenetic proteins (BMPs) are potently proangiogenic; however, the mechanisms underlying the regulation of vessel development by BMPs are not fully understood. To assess the significance of BMP endothelial cell precursor-derived regulator (BMPER) in blood vessel formation in vivo, we investigated its role in retinal angiogenesis. METHODS AND RESULTS In a model of oxygen-ind...

VASCULAR BIOLOGY BMP activity controlled by BMPER regulates the proinflammatory phenotype of endothelium

The endothelium plays a pivotal role in vascular inflammation. Here we study bone morphogenetic protein (BMP) signaling in endothelial inflammation and in particular the role of BMPER, an extracellular BMP modulator that is important in vascular development and angiogenesis. Using the BMP antagonist dorsomorphin or BMP2 as an agonist we show that BMP signaling is essential for the inflammatory ...
